Pharmacological action

Pharmacodynamics：Ivermectin has a better repelling and killing effect on the nematode in vivo and arthropod on body surface.The mechanism of anthelmintic action is promote the presynaptic neuron release the γ- aminobutyric acid,to open the GABA mediated chloride ion channel.Ivermectin has the selectivity and high affinity to the nervous and muscle cell of the invertebrate in the glutamate mediated chloride ion channels of GABA mediate,thus interference the signal transmission between nerve muscle,make the polypide flaccid paralysis,lead the polypide death or excrete to the outside of the body.The inhibition of nematode, site of action is internuncial neuron and excited motor neuron,and the site of action of the arthropod is neuromuscular junction.The expel rate of the haemonchus, osteragia, cooperia, trichostrongyle(include trichostrongylus axei ), strongyle, bunostomum, nematodirus spp, trichocephalus, oesophagostomum radiatum, dictyocaulus of cattle,sheep and chabertia ovina, intestine-sol capsule is 97%-100%.Also have effect on the arthropod,such as fly maggot, acarid and louse.It has less effect on the chewing louse and melophagus ovinus.Ivermectin also has good effect on the tick and breeding flies in faeces,drugs can not kill the ticks immediately,but it can affect the feeding, ecdysis and oviposition then reduce the reproductive capacity.It has the similar effect on the haematobia. The expel rate of the ascaris suum, hyostrongyliasis rubidus, strongyloides ransomi, trichocephalus, oesophagostomum radiatum, metastrongylus, stephanurus dentatus and immaturate polypide is 94%-100%,has good effect on the intestinal trichinella spiralis(invalid to the muscle trichinella spiralis),also have good control action on the haematopinus suis and sarcoptes suis.Invalid to the trematode and tapeworm.

Pharmacokinetics：As the different of the animal species,does form and administration route,the pharmacokinetics of ivermectin has significant difference.The bioavailability of the subcutaneous injection is higher than oral administration,but the absorption of the oral administration is faster than subcutaneous injection.After absorb it can distribute to most organizations well,but it’s not easy enter the cerebrospinal fluid.The apparent volume of distribution of the cattle,sheep and pig is 0.45-2.4L/kg,4.6L/kg and 4L/kg, respectively.Most animal have long half lif,the cattle,sheep and pig is 2-3 days,2-7 days and 0.5 days, respectively.This product metabolism in liver, hydroxylation of cattle and sheep, methylation of pig.Mainly excrete in faeces,less than 5%  prototype or metabolite  excrete in urine.Lactation dam 5% dosage eduction in milk in lactation dam.

Drug interactions：

Used with diethylcarbamazine,may cause the serious or fatal encephalopathy.

Indications:

Macrolides antiparasitic agent.Used to treat the nematodosis, mange and other parasitic insects disease.

Usage and Dosage:

Subcutaneous injection:For one time,Per kg weight,cattle,sheep 0.02ml;pig 0.03ml.

Adverse drug reaction:

1.When treat the cowhide fly larva disease,if the larvae was killed in the key position,may caused serious untoward effect.

2.When inject,the inject site discomfort or transient edema.

Precautions:

1.Lactation period is forbidden.

2.Only used for subcutaneous injection,because intramuscular injection, intravenous injection cause the toxic reaction easily.Each subcutaneous injection point,do not inject over 10ml.

3.The ivermectin injection include the glycerolformal and propylene glycol,only can be used on cattle,sheep and pig.

4.Ivermectin is toxic to shrimp,fish and aquatic organism, residual drugs and package do not polluted water source.

5. Used with diethylcarbamazine,may cause the serious or fatal encephalopathy.
